Introduction
While many types of assays can be performed on the PharmaSeq platform, the current focus of the Company is on genetic testing. To perform a genetic test, two PharmaSeq products are required, a microtransponder-based kit specific for the type of assay and a bench-top instrument (flow reader) that reads the microtransponders in a fluidic format. Custom kits for the specific application can be rapidly developed -- please inquire.
In the authentication and security area, PharmaSeq technology provides a way to tag valuable objects with microtransponders and to instantly read microtransponders when a question arises with regard to the authenticity of an item. To develop an application in this area, two PharmaSeq products are required, microtransponders and an ID reader. Client-specific customization of the ID reader may be also needed -- please inquire.
The size of microtransponders currently available is approximately 500 µm x 500 µm x 100 µm.
Bioassays: Genetic Testing
The flow reader rapidly measures the intensity level of fluorescence emitted by biological molecules bound to the surface of microtransponders while simultaneously reading each microtransponder's electronic ID. Both fluorescence and ID provide sufficient information to determine the presence or absence of mutations in a DNA sample being tested. While there is no particular limit for the number of microtranponders in the assay, currently PharmaSeq supports assays in which up to 1,024 different microtransponders are used.
The system consists of the bench-top flow reader, a computer, and software to control the instrument and analyze the data. The readout phase of the assay typically takes 2-5 min. High accuracy of mutation determination has been reported.
Cystic Fibrosis Mutation Testing
The PharmaSeq cystic fibrosis (CF) mutation assay determines the presence or absence of 51 mutations, including all 24 mutations recommended to be tested for by the American College of Medical Genetics (ACMG). The remaining 27 mutations are grouped into three ethnic panels, specific for Caucasian, Hispanic and African-American populations, thus providing a cost-effective way to determine the mutations for different backgrounds. PharmaSeq provides a select group of key reagents and buffers to facilitate the analysis. This assay is for investigational use only and has not been approved by FDA for clinical testing. Clinical trials are pending.
Microtransponders, Polymer-coated
PharmaSeq provides microtransponders coated with a proprietary polymer layer. The polymer provides hydroxy (OH) and amino (NH2) groups suitable for a variety of methods to conjugate DNA, proteins and organic molecules to the surface of microtransponders. In particular, coated microtransponders can be directly used in automated chemical synthesis of oligonucleotides. Please inquire.
